I've loosened my format since the last time we had one of these threads. Now I've got:

4-Tool
5-Weapon
6-Weapon
7-Weapon
8-Summon (Earlier in the game, rope)

Additional tools are stored (as favorites) along the left side of my inventory and I swap them into #3 and #4 as needed.

I've had to alter my setup as well. It now looks like this:

1-3: Swords
4. Nebula Pickaxe
5. The Axe (still better than the new axes)
6: Ranged weapon
7: Summon
8. Portal Gun
9. Vampire Knives
10. Health potions

Does having health potions on the hotbar help in any way?

I see a lot of people having their pickaxes on their tool bar, as well as torches, why? Do you not use the Shift key at all or do you just find it easier that way?

For me its:

3. Ranged Weapon
4. Safe weapon (like a lance or something with good knockback)
5. Fun weapon

Shifting for me is mega uncomfortable

I do use Auto-Select (Shift) for the pickaxe in many cases, but if I'm going to be digging a long tunnel I want to use Smart Cursor (Ctrl) and pressing both keys at once is too cumbersome.

Just want to give a heads up that smart cursor is toggled.

1 - Panic button. a weapon that requires 0 aiming, and either has ridiculous DPS, or ridiculous knockback. for instance, i am using that new crimson tigerclaw weapon. i have it on Weak modifiers, which reduce its knockback even more. having 70 damage with a use time of 4... it shreds everything, even bosses. will replace with slap-hand if possible.
2 - flying knife. most used weapon, it fits exploration and combat, goes around any obstacle, goes through some walls, penetrates like no tomorrow. overall super versatile.
3 - ranged DPS: megashark.
4 - Ranged groupslayer: dart rifle + crystal darts kills an average of 3 mobs a shot with orichalchum ranger set. plus ricochets keep the petals coming for even MORE damage.
5, 6, 7 - Area Denial: a deadly mix of the Staff of the Frost Hydra, the Nymbus Rod, and that corruption fire wall thingy. i am untouchable in AFK with just this.
8 - Ice Rod: for constricting dangerous enemies like special mimics, making temporary bridges, mining in peace, helping to traverse difficult terrain with hooks, and attacking impunely. i have a Myhtical one :O that thing is awesome ;)
9 - Summons: i love the twins. works for everything.
10 - Mirror Magic, Mirror Mine, Take me Home and Let me Whine.

1: Main Weapon
2: Pickaxe
3-8: Misc stuff (changes depending on what I'm doing)
9: Torches
0: Recall item

1: Melee weapon
2: Ranged weapon
3: Magic weapon
4: Homing weapon
5: Pickaxe
6: Any easily obtained block (Such as mud)
7: Potion 1
8: Potion 2
9: Another weapon of any type
0: Magic mirror
