Most difficult was probably wave 20 of the Frost Moon. Even with post-lunar gear, it takes quite a bit of setup to be able to hit wave 20. You could just use Zenith, but that’s boring and kills the spirit of the challenge.

200 fishing quests is technically the “hardest”, but its hard because of tedium so I don’t count it.
