Shroom biome is largely made by more than the 100+ block needed, and There's no close Corruption/Crimson/Hallow that may overlap , yet there's no Shroom Pylon offered.
Coordinate : 4116 West, 380 Surface

Might want the truffle for that!

The Shroom Pylon can be obtained without Truffle.
It is just a matter of being above a threshold of happiness.
Exemple in screenshot from another save where it actually works with same NPC.

Thinking maybe the parts that haven’t grown in yet are affecting the calculation somehow?

You are too close to the crimson biome nearby in your screenshot (note the crimsand on the left corner):

Moving to the right a bit makes the pylon available:

Though the house not being corrupted would have made it ok, guess i'll nuke even more sand.
