Unfortunately, the truffle is gliched where it takes forever to spawn. Wait a while, and it'll come eventually.

If you want the dryad to move out, try making that house smaller than any of your other npc's houses. For the truffle, the chances are probably about 3% each time day comes. Nice truffle house by the way!
