Might need more mushroom grass. Make sure you have over 100 tiles of grass (grass can be substituted with glowing mushrooms and glowing mushroom trees).

A good rule of thumb I go by is to get the sky dark enough (glowing mushroom biome darkens the sky when you're in it) to the point where it's permanently nighttime where I build the truffles house. Then I absolutely  know i have enough blocks.
